As a neurosurgeon and the chief of Neurosurgical Division, my colleagues and I provide neurosurgical services for various neurosurgical diseases including brain tumors. In brain tumor surgery, maximal safe resection is the gold standard. To achieve maximal resection, we use intraoperative ultrasound, navigation system, and also try to develop fluorescence guided resection technique and real-time imaging for tissue diagnosis, we also hope we can have intraoperative CT and MRI in the near future. To achieve safe excision with minimum deficits, we apply cortical and subcortical mapping, neurophysiological monitoring, awake surgery, functional MRI and tractography navigation… etc., we hope we can co-operate with team or members in neuroscience group to provide better service to our patients and also understand more about the human mind, cognition and behavior. My colleagues and I are interested in the mechanism of photodynamic therapy (PDT) and the clinical applications of PDT for the treatment of cancers, especially malignant gliomas. Recently, we are also interested in photodynamic diagnosis (PDD) and fluorescence guided resection (FGR) of malignant gliomas. We also studied on photodynamic inactivation (PDI) for bacteria, especially multidrug resistant bacteria, and investigated the mechanism of PDI. In cooperation with Prof. Yuan Luo (駱遠), we are developing a fast optical holographic spatial-spectral biomedical imaging technique, which is a novel imaging technique and offers capability to acquire multi-depth resolved images and spectral information of tissue samples. We hope this technique can provide real-time tissue diagnosis during operation in the near future. In cooperation with Prof. Sung-Liang Yu (俞松良), we transfected lentivirus based A549 lung cancer cells harboring luciferase with shRNA library and injected it orthotopically into lung of female NOD/SCID mice. We then pooled the harvested cells from metastatic brain tumors and orthotopically injected again into lung of another mice. By repeated in vivo selection, we found several clones with high incidence of brain metastasis by NGS. Their roles in brain metastasis were characterized and validated in vitro and in vivo.
